چکیده

The phosphatidylcholine specific transfer protein (PCTP) from bovine liver was used to retailor the molecular species composition of phosphatidylcholine (PC) in the membrane of normal (AA) and sickleable (SS) human erythrocytes. Changes in molecular species composition of PC altered morphology as well as cellular deformability and stability as measured with ektacytometry. In normal cells. replacement of native PC with 1 -palmitoyl.2-arachidonoyl PC (PAPC) resulted in a decrease in osmotic fragility with no change in hydration. whereas replacement with 1.2dipalmitoyl PC (DPPC) led to an increased osmotic fragility and cellular hydration. Replacement of native PC by 1 -palmitoyl.2-oleoyl PC (POPC) in normal cells had no

Abnormal phospholipid molecular species of erythrocytes in sickle cell anemia.

As the lipid composition of cell membranes has significant effects upon cellular function, we hypothesized that the membranes of sickle cells might have a distorted lipid composition. Accordingly, we analyzed the molecular species of the choline and ethanolamine glycerophospholipids, the fatty acid composition of the total phospholipids and of the five major individual phospholipids of erythroc...

Abnormal regulation of Mg2+ transport via Na/Mg exchanger in sickle erythrocytes.

Erythrocyte magnesium (Mg2+) deficiency has been demonstrated in sickle cell disease to contribute to erythrocyte dehydration, K loss, and thus sickling. No studies have assessed the functional properties of the Na/Mg exchanger in sickle cell disease. Using Mg(2+)-loaded erythrocytes, we measured Mg2+ efflux induced by extracellular Na+. We estimated that the Na/Mg exchanger had higher maximal ...

An infant Presenting with Cerebrovascular Accident was Diagnosed as a Sickle Cell Disease Patient: a Case Report

Sickle cell disease (SCD) is a known inherited hemoglobin disorder featured by the presence of sickle shaped erythrocytes in the blood. It can cause cerebrovascular accident (CVA) in adults and children and is responsible for the majority of the strokes in children. Repeated blood transfusion are often required in an attempt to dilute blood thus reducing the risk of vaso-occlusion and stroke in...

Static and dynamic rigidities of normal and sickle erythrocytes. Major influence of cell hemoglobin concentration.

Static and dynamic deformabilities of erythrocytes are important determinants of microcirculatory blood flow. To determine the influence of increased cellular hemoglobin concentration on these properties, we quantitated static and dynamic deformabilities of isolated subpopulations of oxygenated normal and sickle erythrocytes with defined cell densities using micromechanical manipulations of ind...
